Thank you for your question. We recommend blocking for one hour at room temperature in 5% nonfat milk + TBST (Blotto A). We also recommend incubating both primary and secondary antibodies in freshly prepared 5% nonfat milk + TBST. To reduce non-specific banding, you can decrease the antibody incubation times to one hour at room temperature for the primary antibody, and 45 minutes at room temperature for the secondary antibody. Wash at least 3 times for 5 minutes each with TBST between antibody incubations. We also strongly recommend including a secondary antibody control (omit primary antibody) to confirm if any of the non-specific bands are from the secondary antibody alone.
